Fast Screeing and Quantitation of Microsystins and Noduralin in Water by Online Injection-UPLC-Orbitrap Mass Spectrometry |

Cyanobacterial blooms can form in warm, slow-moving waters that are rich in nutrients. Typical algal blooms often occur in late summer or early fall. Cyanobacteria produce a wide range of toxic compounds, including microcystins(MC) that are a group of cyclic heptapeptides. WHO(1998) proposed guideline for the most toxic MC-LR as a 1 ㎍/L. For water supplies, fast and accurate analysis of MCs in reservoir or surface water is particularly needed in summer. Conventional methods require manual sample extractions by cartridge to HPLC or LC/MS analysis. In this study, column switching based online preconcentration by EQUAN was combined with high resolution Orbitrap mass spectrometry(Exactive). Targets were MC-RR, MC-YR, MC-LR and noduralin. Filtered sample volume was used 1mL for online injection. Mass resolution of Exactive was set at 50,000 and mass range was 400 - 1100 m/z with 4min run. Practical quantification limit for MC-LR was 0.77 ppb and others were 0.74~1.15 ppb, respectively. For sample application, raw and treated water sample from nationwide 59 drinking water treatment plants (n=118) and Han River samples (n=55) was measured within two days. This method is a very fast, sensitive and accurate technique for the identification and quantitation of MCs in water samples. |
