Giardia lamblia (G. lamblia) is one of the waterborne protozoan parasites and causes Giardiosis. G. lamblia exists in the water system in very low densities and has resistance to environmental stresses. Thereby it is needed to develope a sensitive detection method to detect viable G. lamblia. We developed a reverse transcription (RT)-polymerase chain reaction (PCR) method to detect the mRNA of the heat shock protein (hsp)70 gene for the detection of viable G. lamblia. The results showed that hsp70 mRNA was able to be detected from 100 cyst/100 μL by newly developed RT-PCR. Also, we found the 103-fold increase of the detection sensitivity when hsp70 mRNA was induced by giving heat shock, compared with that of the case that was not subjected to heat shock. In conclusion, by using the RT-PCR method for the hsp70 gene of G. lamblia developed in this study, it was possible to detect viable G. lamblia as well as achieve about 103-fold increase in detection sensitivity. Further studies are needed to detect both viable G. lamblia and Cryptosporidium parvum known to be important waterborne parasites simultaneously by using duplex RT-PCR or real-time RT-PCR. |
