Molecular Characterization of Human Enteroviruses in Korean Surface Water by Integrated Cell Culture RT-PCR |

To investigate the genetic epidemiology of enteroviruses (EVs) in surface waters of Korea, phylogenetic analysis of the 5′ non-coding region (5′ NCR) was conducted. Integrated cell culture RT-PCR with 90 total culturable virus assay-positive cultures yielded 40 positive samples. The results of phylogenetic analysis showed that the nucleotide sequences of EVs could be classified into 4 genetic clusters, and the predominant presence of Korea EVs with great similarities to echoviruses type 30 (76 samples) that might cause aseptic meningitis. Interestingly, two samples in this study had very similar nucleotide sequences to those of coxsackievirus type B1, which was isolated from aseptic meningitis patients in Korea. This might imply that the viral sources possibly had the same genotype and that the virus circulated in both water systems and humans. Three samples formed the same cluster as the nucleotide sequences of coxsackieviruses or echoviruses from China or Japan, and one samples formed the same cluster as that of echovirus from Taiwan. This suggests that EVs in Asia has evolved in a region-specific manner. Further studies are required to obtain more nucleotide sequences of EVs for the continuous control and prevention of their spread and for evolutionary studies. |
