In this study two extraction methods were applied to cultured cyanobacteria and cyanobacteria from Chusori in Daechung reservoir for partitioning microcystins from algae samples. The first extraction method of shaking with extractant was applied to cultured cyanobacteria and the second method using sonifier to cyanobacteria from Chusori in Daechung reservoir. Additional ultrasonication increased the extration efficiencies for microcystins but there were losses of microcystins during SPE pretreatment step.Microcystins extracted from cyanobacteria from Chusori in Daechung reservoir were more than from cultured cyanobacteria by 7-60 folds for different kinds of microcystins. The purities of microcystins were determined by LC/MS. The purities of microcystin RR and LR were 79.5% and 27.3% respectively. This study could offer a basis for producing microcystin standards from cyanobacteria of dam reserviors. If proper seperation and isolation steps are developed the production of microcystin standards will be possible. |
