The microcystin produced according to the standard production procedure (SOP) developed in this study was found to have stability of 2-3 months or more depending on the type in the liquid phase. The pretreatment and refinement procedures were partially improved and the reference materials were re-produced and compared with imported products. As a result of changing the type and capacity of the cartridge for purifying the extract, it was possible to obtain individual components with significantly reduced interfering components compared with the previous year. By changing the sample volume and adjusting the fraction time in the liquid chromatograph and improving the time variable after the fraction, the fraction with improved purity was finally obtained.
As a result, it was confirmed that the purity was much improved than that of the test material produced in the second year, and that it was usable as a reference material for LC-MS/MS. The isomer composition of microcystin produced by bluegreen algae in domestic water system has a pattern of RR >> LR >>> YR. It is considered that the use of the produced reference material in this study was equivalent to the isomer composition of the bluegreen algae generated in Korea, so that it will have a more quantitative performance than the concentration using the imported products.
The calibration properties of K-water's products and the imported reference materials were compared for the major components, RR and LR. The slopes of calibration curve of K-water's products and the imported products for LR were 0.9990 and 0.9993, respectively. The linear slopes of RR were 0.9995 and 0.9980, respectively. The preliminary results applied to the samples were almost similar to those of the two reference materials, so that they can be used for sample quantification sufficiently.
Finaly, microcystin reference material made in K-water were produced with a certificate containing detailed quality specifications.
